Semaglutide Preparation Method

Semaglutide was prepared using a solid-liquid phase synthesis method. The linear peptides Aib8, Arg34GLP-1(7-37), and an adipose side chain acylated agent were prepared by solid-phase synthesis. Under alkaline conditions, the linear peptide Lys26 was modified with the synthesized adipose chain acylated agent to obtain semaglutide. The bioactivity of the prepared sample was determined by time-resolved fluorescence resonance energy transfer immunoassay.
